This month&#8217;s Snapshot straddles precisely that boundary; an image depicting <\/span><b>a key discovery in the regulation of human fertility<\/b><span style=\"font-weight: 300;\">, yet one which, to the artistic eye, evokes the organic growth of <\/span><b>Jean Arp&#8217;s sculptures<\/b><span style=\"font-weight: 300;\">.<\/span><\/p>\n<p><span style=\"font-weight: 300;\">To analyse this snapshot, which forms part of a <\/span><a href=\"https:\/\/www.science.org\/doi\/10.1126\/science.aeb6999\" target=\"_blank\" rel=\"noopener\"><span style=\"font-weight: 300;\">study published in the journal <\/span><i><span style=\"font-weight: 300;\">Science<\/span><\/i><\/a><span style=\"font-weight: 300;\">, we draw on a <\/span><b>combination of artistic and scientific perspectives<\/b><span style=\"font-weight: 300;\">. The soft, abstract forms are reminiscent of the growth or transformation of a living organism. Arp created <\/span><b>forms inspired by nature<\/b><span style=\"font-weight: 300;\"> (leaves, seeds and cells) but without representing them literally. The image on the left shows mice from the control group, while the one of the right shows mice lacking the RANK protein. When we studied RANK in animals we saw that, without it, the reproductive axis is affected and the sex hormones (oestrogen and testosterone) are drastically reduced, impairing fertility in both females and males. Furthermore, we found <\/span><b>mutations in the RANK gene in people with congenital hypogonadotropic hypogonadism<\/b><span style=\"font-weight: 300;\">, a rare condition associated with sexual development and infertility, underlining its importance in human fertility.<\/span><\/p>\n<p><b>E.G.:<\/b><span style=\"font-weight: 300;\"> Another key discovery is that <\/span><b>this process is controlled by the microglia<\/b><span style=\"font-weight: 300;\">, cells from the brain&#8217;s immune system. Apart from eliminating damaged neurons and responding to infection, we&#8217;ve found they have another function: <\/span><b>helping to regulate the neurons that release gonadotropins<\/b><span style=\"font-weight: 300;\"> (GnRH) from the hypothalamus, which are responsible for controlling fertility. Specifically, <\/span><b>RANK controls the microglia activity<\/b><span style=\"font-weight: 300;\">, affecting their communication with these neurons and their function. In short, we&#8217;ve discovered that a protein known for its role in other organs is also essential for controlling the brain system that regulates fertility.<\/span><\/p>\n<p>&nbsp;<\/p>\n<h4><b>Alejandro, what questions has this discovery answered and how did you come up with the answers?<\/b><\/h4>\n<p><b>A:<\/b><span style=\"font-weight: 300;\"> It answers the question of <\/span><b>how the onset of puberty and fertility are regulated in mammals<\/b><span style=\"font-weight: 300;\">. Although we knew that a network of neurons is involved in this process, the role played by the RANK protein was unknown, as was the fact that <\/span><b>the immune system is also involved<\/b><span style=\"font-weight: 300;\">.\u00a0<\/span><\/p>\n<p><span style=\"font-weight: 300;\">A combined approach was used to investigate this. First, we carried out studies on <\/span><b>animal models <\/b><span style=\"font-weight: 300;\">by suppressing the RANK protein (in the whole body or in the microglia) to observe the effects on sexual development and fertility. Next, we analysed the <\/span><b>genetic data from patients with a rare disease that causes infertility<\/b><span style=\"font-weight: 300;\">, identifying mutations in the same gene. And finally, <\/span><b>we studied how the microglia communicate <\/b><span style=\"font-weight: 300;\">with these neurons to understand the mechanism that links the whole system.<\/span><\/p>\n<p>&nbsp;<\/p>\n<h4><b><i>Eva, what impact do you think this discovery will have on society?<\/i><\/b><\/h4>\n<p><b>E.G.:<\/b><span style=\"font-weight: 300;\"> We&#8217;ve identified a new biological mechanism that connects the brain&#8217;s immune system to fertility. <\/span><b>This opens the door to new diagnostic approaches<\/b><span style=\"font-weight: 300;\">, such as in congenital hypogonadotropic hypogonadism, a condition in which half of all cases have no known genetic cause, and in which we detected RANK mutations in 1% of the patients. In the long term, and with further research, this could <\/span><b>help to develop treatments for infertility<\/b><span style=\"font-weight: 300;\"> by modulating the immune system or the RANK pathway. It also paves the way to <\/span><b>explore whether microglia regulate other functions<\/b><span style=\"font-weight: 300;\"> of the hypothalamus, such as appetite, stress or body temperature, or conditions ranging from degenerative diseases to cancer.<\/span><\/p>\n<p>&nbsp;<\/p>\n<h4><b><i>As always, we&#8217;ll finish with a reflection on science and art. Eva, what do you think is the relationship between the two?<\/i><\/b><\/h4>\n<p><b>E.C.: <\/b><span style=\"font-weight: 300;\">Art and science are two sides of the same coin: <\/span><b>humans&#8217; need to understand the world<\/b><span style=\"font-weight: 300;\">. They&#8217;ve always been closely related. It was in the modern era, due to the specialisation of knowledge, when they came to be seen as separate disciplines. Today, that line has become blurred again.
