CNIO<\/p><\/div>\n<h3><b>Dos metres en deu mil\u00b7l\u00e8simes de mil\u00b7l\u00edmetre<\/b><\/h3>\n<p><span style=\"font-weight: 400;\">L\u2019ADN d\u2019una c\u00e8l\u00b7lula humana, desenrotllat, mesura dos metres; hi cap en el nucli de les c\u00e8l\u00b7lules \u2014deu micres de di\u00e0metre\u2014 perqu\u00e8 es plega. En plegar-se es posen en contacte seccions de material gen\u00e8tic molt separades en la cadena d\u2019ADN i aquest contacte \u00e9s el que activa i desactiva <\/span><span style=\"font-weight: 400;\">els <\/span><span style=\"font-weight: 400;\">gens. Els errors en la disposici\u00f3 3D del genoma estan relacionats amb algunes malalties, entre les quals hi ha el c\u00e0ncer.<\/span><\/p>\n<p><span style=\"font-weight: 400;\">\u00abEl plegament de l\u2019ADN determina la forma en qu\u00e8 la c\u00e8l\u00b7lula llegeix, interpreta i mant\u00e9 la informaci\u00f3 en el genoma; el seu estudi est\u00e0 atraient una atenci\u00f3 extraordin\u00e0ria. Tot just comencem a entendre les connexions entre l\u2019organitzaci\u00f3 tridimensional del <\/span><span style=\"font-weight: 400;\">genoma i processos <\/span><span style=\"font-weight: 400;\">clau en l\u2019aparici\u00f3 i progressi\u00f3 del c\u00e0ncer, com la <\/span><span style=\"font-weight: 400;\">reparaci\u00f3 de danys en l\u2019ADN<\/span><span style=\"font-weight: 400;\">\u00bb, assenyalen els investigadors del CNIO <\/span><b>Felipe Cort\u00e9s<\/b><span style=\"font-weight: 400;\">, <\/span><b>Ana Losada <\/b><span style=\"font-weight: 400;\">i <\/span><b>\u00d3scar Fern\u00e1ndez-Capetillo<\/b><span style=\"font-weight: 400;\">, organitzadors del congr\u00e9s juntament amb <\/span><b>Andre Nussenzweig<\/b><span style=\"font-weight: 400;\">, de l\u2019Institut Nacional del C\u00e0ncer dels Estats Units.<\/span><\/p>\n<h3><b>De la <\/b><b>neurotoxicitat<\/b><b> a l\u2019addicci\u00f3 i l\u2019obesitat<\/b><\/h3>\n<p><span style=\"font-weight: 400;\">L\u2019organitzaci\u00f3 del genoma afecta tots els processos de l\u2019organisme, de manera que el congr\u00e9s ha tocat temes diversos: \u00abS\u2019ha parlat sobre per qu\u00e8 la quimioter\u00e0pia pot danyar el nostre cervell o de com es transmet entre generacions l\u2019obesitat. Aquests problemes, tot i que sembli mentida, parteixen de la forma en qu\u00e8 est\u00e0 compactat el genoma i de com s\u2019ho fa la c\u00e8l\u00b7lula per treballar amb aquesta informaci\u00f3\u00bb, diu Fern\u00e1ndez Capetillo.<\/span><\/p>\n<p><span style=\"font-weight: 400;\">La import\u00e0ncia de l\u2019estructura 3D del genoma s\u2019ha descobert de manera gradual. <\/span><a href=\"https:\/\/www.science.org\/doi\/10.1126\/science.1181369?url_ver=Z39.88-2003&amp;rfr_id=ori:rid:crossref.org&amp;rfr_dat=cr_pub%20%200pubmed\"><span style=\"font-weight: 400;\">Noves t\u00e8cniques<\/span><\/a><span style=\"font-weight: 400;\"> desenvolupades en les \u00faltimes d\u00e8cades han anat revelant que hi ha parts llunyanes de la cadena d\u2019ADN que en realitat estan interaccionant.<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Aix\u00f2 ha perm\u00e8s generar \u00abmapes de com s\u2019organitza el genoma, que s\u2019han creuat amb mapes d\u2019on es trenca el genoma \u00bb, explica Cort\u00e9s. \u00abAix\u00ed s\u2019ha vist que \u00e9s m\u00e9s probable que l\u2019ADN es trenqui en les regions en qu\u00e8 \u00e9s estructuralment m\u00e9s fr\u00e0gil. I si mires les mutacions associades al c\u00e0ncer, tamb\u00e9 coincideix\u00bb.<\/span><\/p>\n<h3><b>Organitzaci\u00f3 del genoma i fragilitat de cromosomes<\/b><\/h3>\n<p><span style=\"font-weight: 400;\">Un dels pioners de l\u2019\u00e0rea \u00e9s Andre <\/span><span style=\"font-weight: 400;\">Nussenzweig, amb un treball <\/span><span style=\"font-weight: 400;\">titulat \u00ab<\/span><a href=\"https:\/\/www.cell.com\/cell\/fulltext\/S0092-8674(17)30718-3\"><span style=\"font-weight: 400;\">L\u2019organitzaci\u00f3 del genoma determina la fragilitat dels cromosomes<\/span><\/a><span style=\"font-weight: 400;\">\u00bb. Aquesta descoberta <\/span><span style=\"font-weight: 400;\">va posar Nussenzweig sobre la pista <\/span><span style=\"font-weight: 400;\">d\u2019una \u00e0rea de recerca nova que relaciona les ruptures en el genoma amb l\u2019activitat de les neurones i amb la toxicitat de la quimioter\u00e0pia per al cervell.<\/span><\/p>\n<p><b>Ana Pombo<\/b><span style=\"font-weight: 400;\">, del Max Delbr\u00fcck Center de Berl\u00edn, ha creat una de les t\u00e8cniques que revelen l\u2019estructura 3D del genoma: GAM, sigla en angl\u00e8s del mapatge de l\u2019arquitectura del genoma. Pombo la va desenvolupar el 2017 i ara l\u2019aplica a la investigaci\u00f3 de malalties com l\u2019autisme i l\u2019epil\u00e8psia, i l\u2019efecte de l\u2019<\/span><a href=\"https:\/\/www.mdc-berlin.de\/news\/news\/ana-pombo-art-dna-folding\"><span style=\"font-weight: 400;\">addicci\u00f3 a drogues<\/span><\/a><span style=\"font-weight: 400;\"> sobre les neurones.<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Ana Losada, cap del <\/span><a href=\"https:\/\/www.cnio.es\/investigacion-e-innovacion\/programas-cientificos\/programa-de-oncologia-molecular\/grupo-de-dinamica-cromosomica\/\"><span style=\"font-weight: 400;\">grup de Din\u00e0mica Cromos\u00f2mica del CNIO<\/span><\/a><span style=\"font-weight: 400;\">, ha presentat en el congr\u00e9s el seu treball amb la <\/span><a href=\"https:\/\/www.cnio.es\/noticias\/investigadores-del-cnio-ayudan-a-entender-como-funciona-la-proteina-que-hace-lazos-de-adn-en-el-genoma-humano\/\"><span style=\"font-weight: 400;\">cohesina<\/span><\/a><span style=\"font-weight: 400;\">, la prote\u00efna que fa possible el plegament del genoma <\/span><span style=\"font-weight: 400;\">formant <\/span><span style=\"font-weight: 400;\">lla\u00e7os d\u2019ADN.
