Aquestes noves \u201ctisores gen\u00e8tiques\u201d&#8221; han suposat una aut\u00e8ntica revoluci\u00f3 en la biologia i la medicina, i entre d\u2019altres aplicacions, tenen el potencial de desenvolupar noves ter\u00e0pies contra el c\u00e0ncer i curar malalties gen\u00e8tiques heredit\u00e0ries.<\/p>\n<p>No obstant aix\u00f2, aquesta eina d\u2019edici\u00f3 gen\u00e8tica no \u00e9s l\u2019\u00fanica. De fet \u2013sense anar m\u00e9s lluny\u2013 un equip espanyol del <a href=\"https:\/\/www.upf.edu\/web\/synbio\" target=\"_blank\" rel=\"noopener noreferrer\">Grup de Recerca en Biologia Sint\u00e8tica Translacional<\/a> de la UPF est\u00e0 desenvolupant una nova tecnologia anomenada Uni-large que podria tenir fins i tot m\u00e9s \u00e8xit que la guardonada CRISPR-Cas9, ja que es m\u00e9s universal, efica\u00e7 i segura. En aquest article entrevistem Marc G\u00fcell, cap del grup de Biologia Sint\u00e8tica Translacional de la UPF, qui lidera aquest projecte. <a href=\"https:\/\/caixaimpulse.com\/en-US\/web\/guest\/projects\/-\/caixaimpulse\/project\/254148\" target=\"_blank\" rel=\"noopener noreferrer\">Uni-large<\/a> t\u00e9 el suport de CaixaImpulse, un programa de la Fundaci\u00f3 \u201dla Caixa\u201d en Investigaci\u00f3 i Salut que impulsa la transfer\u00e8ncia de la recerca a la societat. A continuaci\u00f3 G\u00fcell ens descobreix alguns dels secrets que hi ha al darrere d\u2019aquesta prometedora t\u00e8cnica.<\/p>\n<p><strong>Uni-large, unes prometedores \u201ctisores gen\u00e8tiques\u201d<\/strong><\/p>\n<p><em>En primer lloc, qu\u00e8 \u00e9s Uni-large?<\/em><\/p>\n<p>Uni-large \u00e9s una nova tecnologia d&#8217;edici\u00f3 gen\u00e8tica que ens permet tallar i enganxar ADN amb precisi\u00f3 i efic\u00e0cia, a m\u00e9s amb la propietat que podem enganxar trossos d&#8217;ADN que siguin molt llargs. \u00c9s un tipus de &#8220;tisores gen\u00e8tiques&#8221;, tal com es coneix popularment a les eines d&#8217;edici\u00f3 gen\u00e8tica.<\/p>\n<p><em>I qu\u00e8 es pot aconseguir amb aquestes &#8220;tisores gen\u00e8tiques&#8221;?<\/em><\/p>\n<p>Les aplicacions de l\u2019edici\u00f3 gen\u00e8tica s\u00f3n infinites i apliquen a diferents disciplines de la bioenginyeria. En l\u2019\u00e0mbit terap\u00e8utic, ens permeten reprogramar \u201cerrors\u201d en el codi gen\u00e8tic que causen malalties: podem tallar una seq\u00fc\u00e8ncia d\u2019ADN i substituir-la per una altra en bon estat, amb la finalitat que recuperi la seva funci\u00f3 normal i la malaltia remeti.<\/p>\n<p><em>En qu\u00e8 es diferencia Uni-large d\u2019altres eines d\u2019edici\u00f3 gen\u00e8tica com CRISPR-Cas9?<\/em><\/p>\n<p>D\u2019una banda, \u00e9s m\u00e9s universal, ja que mentre que el m\u00e8tode CRISPR corregeix mutacions individuals de cada pacient, Uni-large t\u00e9 el potencial d\u2019incloure tots els pacients que pateixen una determinada malaltia. D\u2019altra banda, CRISPR talla el genoma i fa servir la reparaci\u00f3 cel\u00b7lular de l\u2019ADN per a l\u2019edici\u00f3, fet que pot resultar impredictible. Nosaltres, en canvi, fem un control m\u00e9s estricte de l\u2019edici\u00f3 mitjan\u00e7ant un element extra que sempre \u201ctalla i enganxa\u201d de la mateixa manera, aix\u00ed que \u00e9s m\u00e9s segur. Finalment, Uni-large \u00e9s m\u00e9s efica\u00e7 que altres \u201ctisores gen\u00e8tiques\u201d amb gens molt grans, cosa que potencialment pot ajudar-nos a curar malalties heredit\u00e0ries amb aquestes caracter\u00edstiques.<\/p>\n<p><em>En podries posar un exemple?<\/em><\/p>\n<p>Ara mateix estem fent servir Uni-large per la distr\u00f2fia muscular causada pel d\u00e8ficit de merosina. Aquesta malaltia es produeix per una mutaci\u00f3 en el gen LAMA2. Tot i ser la distr\u00f2fia muscular cong\u00e8nita m\u00e9s freq\u00fcent, per ara no t\u00e9 tractament, aix\u00ed que el nostre objectiu \u00e9s poder curar-la.<\/p>\n<p><em>I quin potencial de futur t\u00e9 aquesta t\u00e8cnica?<\/em><\/p>\n<p>Partint de la premissa que alguns animals, formats amb els mateixos constituents biol\u00f2gics que tu i jo, s\u00f3n molt m\u00e9s resistents a la radiaci\u00f3 c\u00f2smica o al c\u00e0ncer, o fins i tot uns pocs poden viure perp\u00e8tuament, l&#8217;enginyeria de sistemes biol\u00f2gics realment t\u00e9 propietats il\u00b7limitades.
