A Espanya, hi ha cinc casos per cada 100.000 habitants. Aquesta malaltia est\u00e0 causada per una mutaci\u00f3 que provoca que hi hagi baixos nivells de la prote\u00efna frataxina. Malgrat que encara no es coneix gaire b\u00e9 la funci\u00f3 d&#8217;aquesta prote\u00efna, est\u00e0 relacionada amb la regulaci\u00f3 dels nivells de ferro. Els s\u00edmptomes de l&#8217;AF es comencen a manifestar durant l&#8217;adolesc\u00e8ncia. La manca de frataxina afecta diferents tipus de neurones i provoca dificultats en la coordinaci\u00f3 dels moviments dels pacients. Aquesta malaltia causa en els afectats una p\u00e8rdua progressiva de moltes de les funcions necess\u00e0ries per a una autonomia personal: p\u00e8rdua de sensibilitat, descoordinaci\u00f3 en els moviments, immunodefici\u00e8ncia i, en molts casos, diabetis i problemes card\u00edacs greus.<\/p>\n<p style=\"text-align: justify;\"> L&#8217;AF \u00e9s una malaltia recessiva autosom\u00e0tica, que vol dir que el pacient ha d&#8217;heretar dos gens afectats, un de cadascun dels pares, perqu\u00e8 la malaltia es manifesti. Una persona que t\u00e9 nom\u00e9s una c\u00f2pia anormal d&#8217;un gen per a una malaltia gen\u00e8tica recessiva es denomina portador. Un portador no desenvolupar\u00e0 la malaltia per\u00f2 pot transmetre el gen afectat als seus fills. Si ambd\u00f3s pares s\u00f3n portadors del gen de l&#8217;AF, els seus fills tindran un 25 % de possibilitats de tenir la malaltia i un 50 % de probabilitat d&#8217;heretar un gen anormal que ells, al seu torn, podrien passar als seus fills.<\/p>\n<p style=\"text-align: justify;\"> <strong>Recerca actual<\/strong><\/p>\n<p style=\"text-align: justify;\"> <strong><img decoding=\"async\" alt=\"\" src=\"https:\/\/blog.caixaresearch.org\/wp-content\/uploads\/2020\/04\/ca\/gate2brain2.png\" style=\"width: 400px; height: 361px;\"\/> <\/strong><\/p>\n<p style=\"text-align: justify;\"> Actualment hi ha ter\u00e0pies de reempla\u00e7ament proteic per tractar malalties caracteritzades per la manca d&#8217;una prote\u00efna espec\u00edfica. L&#8217;exemple m\u00e9s conegut \u00e9s el de la insulina per tractar la diabetis. En el cas de l&#8217;AF, el tractament amb frataxina no es podria aplicar directament ja que la barrera hematoencef\u00e0lica (BHE) impedeix l&#8217;acc\u00e9s del 98 % de les subst\u00e0ncies al cervell per protegir-lo. Per obtenir una ter\u00e0pia de reempla\u00e7ament per l&#8217;AF, la BHE \u00e9s el primer obstacle que cal superar. El nostre laboratori ha treballat durant molt de temps en el desenvolupament de llan\u00e7adores pept\u00eddiques, que s\u00f3n compostos capa\u00e7os d&#8217;aprofitar els mecanismes naturals de transport del cervell per augmentar el pas de subst\u00e0ncies que no ho poden fer per elles mateixes. Aquests p\u00e8ptids llan\u00e7adora han demostrat que s\u00f3n \u00fatils per augmentar el transport de diferents tipus de compostos tant en models cel\u00b7lulars com en models de ratol\u00ed.<\/p>\n<p style=\"text-align: justify;\"> El projecte Gate2Brain, en el marc de la convocat\u00f2ria RecerCaixa, ha perm\u00e8s explorar la preparaci\u00f3 de diversos derivats de frataxina modificats amb diferents llan\u00e7adores pept\u00eddiques. La nostra experi\u00e8ncia en el camp de la modificaci\u00f3 de prote\u00efnes i l&#8217;\u00fas de p\u00e8ptids llan\u00e7adora ha perm\u00e8s assajar aquests compostos en models cel\u00b7lulars.
