Aquest va ser el tema del <\/span><a href=\"https:\/\/www.cnio.es\/eventos\/structural-and-molecular-biology-of-the-dna-damage-response\/\" rel=\"noopener noreferrer\" target=\"_blank\">CNIO-\u201dla Caixa\u201d Frontiers Meeting<\/a> del passat mes de maig, un esdeveniment que va reunir a Madrid investigadors d\u2019arreu del m\u00f3n l\u00edders en aquest camp.<\/p>\n<p dir=\"ltr\"> <span id=\"docs-internal-guid-e9fc3de9-7fff-9abd-2bd2-992e17f02c77\">A l\u2019interior de les c\u00e8l\u00b7lules que formen els teixits del cos hum\u00e0 tenen lloc multitud de processos d\u2019enorme complexitat, des de la protecci\u00f3 enfront de pat\u00f2gens fins a la c\u00f2pia i reparaci\u00f3 de danys del nostre material gen\u00e8tic. La major part d\u2019aquestes activitats la fan grans complexos macromoleculars on un conjunts de prote\u00efnes s\u2019associa per fer, com si fossin m\u00e0quines, tasques de diversa \u00edndole.\u00a0<\/span><\/p>\n<p dir=\"ltr\"> S\u00f3n d\u2019especial inter\u00e8s els complexos macromoleculars que mantenen la integritat del genoma. El nostre ADN cont\u00e9 tota la informaci\u00f3 necess\u00e0ria per al correcte funcionament cel\u00b7lular, per\u00f2 aquesta ha de ser llegida (transcripci\u00f3) i copiada (reproducci\u00f3) sense errors. A m\u00e9s, aquesta informaci\u00f3 pateix constantment danys generats per l\u2019exposici\u00f3 a agents de l\u2019ambient (f\u00edsics com radiacions, i qu\u00edmics com el tabac), com tamb\u00e9 productes del metabolisme de les c\u00e8l\u00b7lules.<\/p>\n<p dir=\"ltr\"> <span id=\"docs-internal-guid-e9fc3de9-7fff-9abd-2bd2-992e17f02c77\">Per sort, disposem d\u2019una xarxa de complexos macromoleculars capa\u00e7os de recon\u00e8ixer el dany a l\u2019ADN i corregir-lo. No obstant aix\u00f2, quan fallen, fonamentalment perqu\u00e8 les prote\u00efnes contenen una mutaci\u00f3 que impedeix el funcionament normal, els errors s\u2019acumulen i poden desembocar en c\u00e0ncer i altres malalties. De fet, la manca d\u2019una correcta reparaci\u00f3 dels danys al genoma \u00e9s un pas necessari per a la superviv\u00e8ncia del tumor.<\/span><\/p>\n<p dir=\"ltr\"> <span id=\"docs-internal-guid-e9fc3de9-7fff-9abd-2bd2-992e17f02c77\">Un exemple de la import\u00e0ncia d\u2019aquests sistemes de protecci\u00f3 de l\u2019ADN i de com poden inspirar noves ter\u00e0pies \u00e9s el cas de les prote\u00efnes BRCA1 i BRCA2, que tenen una gran rellev\u00e0ncia en la predisposici\u00f3 al c\u00e0ncer de mama i al d\u2019ovari familiar. BRCA1 i BRCA2 s\u00f3n dues prote\u00efnes que participen en la resposta de la c\u00e8l\u00b7lula davant lesions a l\u2019ADN. Gr\u00e0cies al fet que sabem que les c\u00e8l\u00b7lules amb mutacions BRCA1 i BRCA2 s\u00f3n especialment sensibles a la inhibici\u00f3 de PARP, un enzim implicat en la reparaci\u00f3 de l\u2019ADN, s\u2019han aprovat inhibidors de la prote\u00efna PARP per al tractament d\u2019alguns tipus de tumors de mama i ovari.<\/span><\/p>\n<p dir=\"ltr\"> <span id=\"docs-internal-guid-e9fc3de9-7fff-9abd-2bd2-992e17f02c77\">Els beneficis de comprendre els detalls de la maquin\u00e0ria cel\u00b7lular encarregada de copiar i tenir cura del nostre genoma s\u00f3n molts, per\u00f2 fer-ho \u00e9s una tasca extraordin\u00e0riament complicada. Aquests complexos macromoleculars es troben en quantitats petites, i, a m\u00e9s les estructures que els formen s\u00f3n flexibles i es mouen com a part del seu propi mecanisme d\u2019acci\u00f3.<\/span><\/p>\n<p dir=\"ltr\"> <img decoding=\"async\" alt=\"\" src=\"https:\/\/blog.caixaresearch.org\/wp-content\/uploads\/2020\/04\/ca\/propuesta_grafico_400p.jpg\" style=\"width: 400px; height: 396px;\"\/><br \/> <span style=\"font-size:11px;\">Figura cedida per:\u00a0Rafael Fern\u00e1ndez-Leiro i \u00d3scar Llorca<\/span><\/p>\n<p dir=\"ltr\"> <span id=\"docs-internal-guid-e9fc3de9-7fff-9abd-2bd2-992e17f02c77\">Tradicionalment, per poder estudiar-los era necessari obtenir grans quantitats de les prote\u00efnes implicades i aconseguir que formessin vidres per despr\u00e9s poder analitzar mitjan\u00e7ant t\u00e8cniques de difracci\u00f3 de raigs X. Per\u00f2 aquestes metodologies presenten limitacions quan es tracta de grans complexos macromoleculars o de prote\u00efnes molt flexibles i din\u00e0miques.\u00a0<\/span><\/p>\n<p dir=\"ltr\"> <span id=\"docs-internal-guid-e9fc3de9-7fff-9abd-2bd2-992e17f02c77\">Durant els \u00faltims anys, una altre t\u00e8cnica, la criomicrosc\u00f2pia (crio-ME) ha revolucionat el m\u00f3n de la biologia. Mitjan\u00e7ant crio-ME podem ara observar directament grans complexos macromoleculars fent servir quantitats relativament modestes de material i determinar-ne l\u2019estructura tridimensional amb detall at\u00f2mic. I, amb aquesta visi\u00f3 de detall, han sorgit tamb\u00e9 noves preguntes. La revoluci\u00f3 \u00e9s de tal magnitud que els investigadors que van liderar aquests desenvolupaments van compartir el premi Nobel de qu\u00edmica el 2017.<\/span><\/p>\n<p dir=\"ltr\"> <span id=\"docs-internal-guid-e9fc3de9-7fff-9abd-2bd2-992e17f02c77\">Vivim un moment excitant, i per aix\u00f2 el CNIO \u201cla Caixa\u201d Frontiers Meeting, celebrat al maig d\u2019enguany, va estar impregnat d\u2019entusiasme i fascinaci\u00f3 pels aven\u00e7os aconseguits en tan poc temps.
