Era el 8 de desembre de 2020 i la biomedicina establia un r\u00e8cord hist\u00f2ric. En comparaci\u00f3, el desenvolupament i l\u2019aprovaci\u00f3 de la vacuna de l\u2019ebola va requerir 5 anys; la del xarampi\u00f3, 7. Aquesta velocitat sense precedents va ser possible gr\u00e0cies a una combinaci\u00f3 de factors \u2014entre d\u2019altres, un finan\u00e7ament i una cooperaci\u00f3 global excepcionals\u2014, per\u00f2 n\u2019hi va haver un que va marcar la gran difer\u00e8ncia: <\/span><b>la tecnologia de l\u2019ARN<\/b><span style=\"font-weight: 400;\">.\u00a0<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Lluny de ser una soluci\u00f3 puntual, aquest aven\u00e7 ha obert les portes a una nova generaci\u00f3 de ter\u00e0pies: les ter\u00e0pies basades en ARN \u2014una mol\u00e8cula clau en processos essencials, com la s\u00edntesi de prote\u00efnes\u2014, que estan revolucionant la medicina ja que ofereixen formes m\u00e9s vers\u00e0tils, precises i personalitzades de combatre malalties.\u00a0<\/span><\/p>\n<p><span style=\"font-weight: 400;\">\u00ab<\/span><b>Les vacunes d\u2019ARN nom\u00e9s s\u00f3n el primer pas d\u2019una revoluci\u00f3.<\/b><span style=\"font-weight: 400;\"> En un futur pr\u00f2xim veurem no tan sols noves vacunes, sin\u00f3 tamb\u00e9 f\u00e0rmacs que, utilitzant una tecnologia similar, corregiran o milloraran diferents malalties\u00bb, explica <\/span><a href=\"https:\/\/caixaresearch.org\/es\/convocatoria-caixaresearch-investigacion-salud-2024-proyecto-terapias-cancer-higado\"><span style=\"font-weight: 400;\">Puri Fortes<\/span><\/a><span style=\"font-weight: 400;\">, investigadora CaixaResearch del <\/span><a href=\"https:\/\/cima.cun.es\/investigacion\/personal-investigacion\/purificacion-fortes-alonso\"><span style=\"font-weight: 400;\">CIM de la Universidad de Navarra<\/span><\/a><span style=\"font-weight: 400;\"> especialitzada en noves ter\u00e0pies d\u2019ARN per tractar el c\u00e0ncer de fetge.\u00a0<\/span><\/p>\n<p><b>El potencial \u00e9s enorme, per\u00f2 tamb\u00e9 ho s\u00f3n els desafiaments:<\/b><span style=\"font-weight: 400;\"> aconseguir que l\u2019ARN sigui m\u00e9s estable en l\u2019organisme, minimitzar els efectes adversos, dirigir-lo amb precisi\u00f3 als teixits afectats o perfeccionar els sistemes d\u2019administraci\u00f3.\u00a0<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Amb l\u2019ajut de cinc investigadors de la xarxa CaixaResearch, explorem com aquesta nova generaci\u00f3 de ter\u00e0pies, fruit de d\u00e8cades d\u2019investigaci\u00f3 silenciosa, podria marcar <\/span><b>l\u2019inici d\u2019una medicina m\u00e9s r\u00e0pida, m\u00e9s precisa i m\u00e9s personalitzada<\/b><span style=\"font-weight: 400;\">.\u00a0<\/span><\/p>\n<h2><b>M\u00e9s enll\u00e0 de les vacunes<\/b><\/h2>\n<p><span style=\"font-weight: 400;\">L\u2019ARN (\u00e0cid ribonucleic) \u00e9s un parent proper de l\u2019ADN i existeix en diferents formes. El m\u00e9s conegut \u00e9s l\u2019<\/span><b>ARN missatger (ARNm)<\/b><span style=\"font-weight: 400;\">, que copia les instruccions gen\u00e8tiques de l\u2019ADN i les transporta fins als ribosomes, on es <\/span><b>fabriquen les prote\u00efnes essencials per a la vida<\/b><span style=\"font-weight: 400;\">. Aquest \u00e9s un dels avantatges m\u00e9s prometedors de la ter\u00e0pia basada en ARN\u00bb, assenyala Portero.\u00a0<\/span><\/p>\n<h2><b>El sistema immune com a aliat<\/b><\/h2>\n<p><span style=\"font-weight: 400;\">L\u2019ARN tamb\u00e9 est\u00e0 revolucionant el tractament del c\u00e0ncer, especialment a trav\u00e9s de les immunoter\u00e0pies i les vacunes antitumorals. En aquest context tindr\u00e0 un paper decisiu el <\/span><a href=\"https:\/\/fundacionlacaixa.org\/ca\/caixaresearch-institute\"><span style=\"font-weight: 400;\">CaixaResearch Institute<\/span><\/a><span style=\"font-weight: 400;\">, el primer centre de recerca especialitzat en immunologia d\u2019Espanya i un dels primers a Europa. \u00ab<\/span><b>L\u2019enfocament pioner del CaixaResearch Institute \u00e9s fonamental<\/b><span style=\"font-weight: 400;\">, ja que la immunologia est\u00e0 implicada en la majoria de patologies cr\u00f2niques, com tamb\u00e9 en l\u2019envelliment i les malalties que duu associades. Els seus aven\u00e7os es poden aplicar a moltes \u00e0rees de la salut <\/span><span style=\"font-weight: 400;\">de manera transversal<\/span><span style=\"font-weight: 400;\">. Per exemple, <\/span><span style=\"font-weight: 400;\">aquesta transversalitat l\u2019apliquem en <\/span><span style=\"font-weight: 400;\">els nostres estudis sobre immunologia tumoral <\/span><span style=\"font-weight: 400;\">ja que els mateixos mecanisme poden ser rellevants<\/span><span style=\"font-weight: 400;\"> en malalties infeccioses, autoimmunes o neuroimmunol\u00f2giques\u00bb, explica Celi\u00e0-Terrassa.\u00a0<\/span><\/p>\n<h2><b>Noves instruccions contra el c\u00e0ncer<\/b><\/h2>\n<p><span style=\"font-weight: 400;\">A m\u00e9s de l\u2019ARN missatger, hi ha altres tipus d\u2019ARN amb un gran potencial terap\u00e8utic. \u00c9s el cas dels tipus d\u2019<\/span><b>ARN no codificants, que no participen en la fabricaci\u00f3 de prote\u00efnes, per\u00f2 s\u00ed en la regulaci\u00f3 de m\u00faltiples processos cel\u00b7lulars<\/b><span style=\"font-weight: 400;\">. Molts d\u2019aquests ARN s\u00f3n presents a les c\u00e8l\u00b7lules tumorals i poden marcar la difer\u00e8ncia entre un tumor benigne i un de maligne. Si apliques radioter\u00e0pia o quimioter\u00e0pia a aquests tumors, trenques el seu ADN, per\u00f2 sobreviuen gr\u00e0cies al NIHCOLE, que actua com una eina de reparaci\u00f3 molt potent\u00bb, aclareix Puri Fortes. Per aquest motiu, l\u2019objectiu \u00e9s clar: \u00ab<\/span><b>Volem atacar el NIHCOLE per desactivar la maquin\u00e0ria que permet al tumor reparar el seu ADN<\/b><span style=\"font-weight: 400;\">. Si no el pot reparar, la c\u00e8l\u00b7lula tumoral no pot sobreviure\u00bb.\u00a0<\/span><\/p>\n<p><span style=\"font-weight: 400;\">A m\u00e9s del fetge, l\u2019ARN NIHCOLE tamb\u00e9 s\u2019ha identificat en altres tipus de c\u00e0ncer, com els de mama, pulm\u00f3, c\u00f2lon o cap i coll. \u00ab<\/span><b>El potencial d\u2019aquestes ter\u00e0pies \u00e9s enorme.<\/b><span style=\"font-weight: 400;\"> Ens cal descobrir m\u00e9s ARN com aquest: con\u00e8ixer-ne les seq\u00fc\u00e8ncies, entendre com es pleguen en una estructura cel\u00b7lular i quina funci\u00f3 tenen. Aix\u00ed podrem fer el cam\u00ed al rev\u00e9s i <\/span><b>obtenir milers de possibles ter\u00e0pies de manera molt m\u00e9s \u00e0gil i senzilla<\/b><span style=\"font-weight: 400;\">\u00bb, assenyala Fortes.\u00a0<\/span><\/p>\n<h2><b>Estrat\u00e8gies de precisi\u00f3 per a malalties <\/b><b>hep\u00e0tiques<\/b><\/h2>\n<p><span style=\"font-weight: 400;\">Entre els ARN no codificants amb m\u00e9s potencial terap\u00e8utic destaca l\u2019ARN interferent (siRNA) o <\/span><b>ARN antisentit, capa\u00e7 de silenciar gens espec\u00edfics<\/b><span style=\"font-weight: 400;\">. Tot i que \u00e9s habitual investigar-ne l\u2019\u00fas en ter\u00e0pies contra el c\u00e0ncer, aquesta eina tamb\u00e9 mostra un gran potencial en el cas de malalties rares. L\u2019equip de <\/span><a href=\"https:\/\/caixaresearch.org\/ca\/convocatoria-caixaimpulse-innovacio-salut-2023-projecte-hepatiques-arn-terapeutic\"><span style=\"font-weight: 400;\">Malu Mart\u00ednez-Chantar<\/span><\/a><span style=\"font-weight: 400;\">, investigadora principal del <\/span><a href=\"https:\/\/www.cicbiogune.es\/people\/mlmartinez\"><span style=\"font-weight: 400;\">Laboratori de Malalties Hep\u00e0tiques del CIC bioGUNE<\/span><\/a><span style=\"font-weight: 400;\"> que t\u00e9 el suport de <\/span><a href=\"https:\/\/www.cicbiogune.es\/news\/la-fundacion-la-caixa-impulsa-dos-proyectos-biomedicos-de-cic-biogune-para-trasladar-sus\"><span style=\"font-weight: 400;\">CaixaImpulse Innovaci\u00f3<\/span><\/a><span style=\"font-weight: 400;\">, estudia com es pot aplicar aquesta tecnologia per<\/span><b> bloquejar un gen implicat en el metabolisme cel\u00b7lular<\/b><span style=\"font-weight: 400;\">.
